It means "your greatest strength".

"What are your greatest strengths and weaknesses?"
"Quelles sont vos principales qualités et vos principaux défauts ?"

That's a scary question recruiters like to ask.

"votre principale qualité" = "your main quality"
That's something you often get asked about when you apply for a job. "Quelle est votre principale qualité ?" --> "What is your main quality?"

a "défaut" is a weakness.

So it means "your greatest weakness".

Pay attention to the spelling, though. Votre principal (no e) défaut. Défaut is masculine.
